Developing staining protocols for visualization of tissue-engineering scaffolds using micro computed tomography in native wet state.

چکیده
BNC-alginate and silk fibroin tissue-engineering scaffolds were stained with X-ray contrast agents in order to visualize internal microstructure in the native wet state with microcomputed tomography. A successful protocol employing amphiphilic contrast agents (CAs) dissolved in a waterbased staining solution was used. The CAs were then fixed to the scaffold by neutralizing their charged functional groups, increasing their hydrophobicity and retention on the scaffold surface in water. While some unresolved issues concerning homogeneous staining and strength of contrast remain, these first successes constitute an important milestone by identifying good contrast agent candidates and staining protocols for longitudinal monitoring of tissue-engineering studies.

Evaluation of Effect of Different Computed Tomography Scanning Protocols on Hounsfield Unit and Its Impact on Dose Calculation by Treatment Planning System
Introduction: In radiotherapy treatment planning system (TPS), basic input is the data from computed tomography (CT) scan, which takes into account the effect of inhomogeneities in dose calculations. Measurement of CT numbers may be affected by scanner-specific parameters. Therefore, it is important to verify the effect of different CT scanning protocols on Hounsfield unit (HU) and its impact o...
متن کاملMicro-computed tomographical imaging of soft biological materials using contrast techniques.
The aim of this work was to introduce high-resolution computed tomography (micro-CT) for scaffolds made from soft natural biomaterials, and to compare these data with the conventional techniques scanning electron microscopy and light microscopy. Collagen-based scaffolds were used as examples. Unlike mineralized tissues, collagen scaffolds do not provide enough X-ray attenuation for micro-CT ima...
متن کاملComparative evaluation of nanofibrous scaffolding for bone regeneration in critical-size calvarial defects.
In a previous study we found that nanofibrous poly(l-lactic acid) (PLLA) scaffolds mimicking collagen fibers in size were superior to solid-walled scaffolds in promoting osteoblast differentiation and bone formation in vitro. In this study we used an in vivo model to confirm the biological properties of nanofibrous PLLA scaffolds and to evaluate how effectively they support bone regeneration ag...
متن کاملStrategy of computed tomography image optimization in cervical vertebra and neck soft tissue in emergency patients
Introduction: Due to the use of ionizing radiation in the CT scan, optimal parameters should be used to reduce the risk of cancer in patients who are constantly exposed to X-rays. The aim of this study was to optimize the parameters used in CT scan of cervical vertebrae and neck soft tissue with minimal loss of image quality Materials and Methods: In this s...
متن کاملHeidelberg-mCT-Analyzer: a novel method for standardized microcomputed-tomography-guided evaluation of scaffold properties in bone and tissue research
Bone tissue engineering and bone scaffold development represent two challenging fields in orthopaedic research. Micro-computed tomography (mCT) allows non-invasive measurement of these scaffolds' properties in vivo. However, the lack of standardized mCT analysis protocols and, therefore, the protocols' user-dependency make interpretation of the reported results difficult. To overcome these issu...
